DUI Task Force Report followup. The Criminal Practice Subcommittee of the House Judiciary Committee deferred to its summer study sessions just about all of the work product of the Governor's DUI Task Force. As you will recall, the task force recommended repeal of the litter pickup penalty provision and several other items on the prosecutorial community's wish list. Next on the list were, administrative revocations of driver's licenses for DUI offenders, and expansion of our open container prohibition, and some mandatory use of ignition interlock devices for repeat offenders. For the treatment community, this last item gives pause for concern because of the fiscal effect of taking funding away from treatment resources in search of an unproven silver bullet in the form of ignition interlock devices. These legislative study committee sessions will be the perfect opportunity educate our state policy makers that the state's Alcohol and Drug Addiction Treatment fund has been the only source of any growth in treatment resource funding since its enactment in 1999. Election update: Kevin Gallagher will face Jeanne Richardson in the Democratic Primary election on May 31 to determine who will succeed Beverly Marrero as State Representative for District 89, which covers the Midtown area of Memphis.
Inhibition of nuclear factor-kappa B NF-B ; is a therapeutic target for inducing apoptosis of a variety of tumor cells 13, 40 ; . The NF-B family of proteins consists of five members: c-Rel, p65 RelA, RelB, p50 p105 NF-B1 ; , and p52 p100 NF-B2 ; 5 ; . NF-B can be activated by a variety of stimuli, including interleukin-1 IL-1 ; and TNF, and among its multiple effects, it induces anti-apoptotic proteins and promotes cell survival 17 ; . There are two NF-B pathways, the canonical and the noncanonical pathways. In the canonical NF-B pathway, NF-B is retained in the cytosol by its.
Q Elliot Wilbur : Hi, Elliot Wilbur, CIBC World Markets. Wanted to ask a line of questioning around expected RESTASIS performance in 2007. I know last quarter, you talked about repositioning the sales force call activity to reignite volume in that product, and I guess just looking at the RXs, it doesn't quite seem like we have yet hit the inflection point that you would be thinking about, in, at least in terms of hitting the top end of the range for '07. So David, I was just wondering if you could comment on that a little bit in terms of whether or not you think there is still a little bit more work to do there, or you think that things are basically tracking where you want them to. And also, in terms of the implied growth rate, 18 to 25%, I assuming it would be fair to include some sort of price increase in there as well? A : Yeah. Q Elliot Wilbur : Okay. A David Pyott : Well. First of all, so far, we haven't had a price increase in 2007, normally we do. Then if I look at the Verispan data which we have, and we don't buy IMS data, there is an upward trend. If I look at say the fourth quarter in terms of NRXs, versus the third, we did increase quarter-over-quarter, Q4 versus Q3 plus 4%. That is a better trend than we have had from Q3 to Q2, which was actually a minus 3%, which certainly sounded my alarm bells, and got people going around here. And then I think really answering your question centrally, as you know, when you put on basically double the amount of capacity, it takes a couple of months for the effects of all of those calls to translate into prescribing activity. And so our ophthalmology organization is all over this. And I think we also have the benefit of stability because at the beginning of the last year when you add a lot of people there is always a certain amount of dislocation, and I think the organization has really settled down, and they know this is one of their top priorities for 2007. This annual report and other written reports and oral statements made from time to time by the Company may contain so-called "forward-looking statements, " all of which are subject to risks and uncertainties. One can identify these forward-looking statements by their use of words such as "expects, " "plans, " "will, " "estimates, " "forecasts, " "projects" and other words of similar meaning. One can also identify them by the fact that they do not relate strictly to historical or current facts. These statements are likely to address the Company's growth strategy, financial results, product approvals and development programs. One must carefully consider any such statement and should understand that many factors could cause actual results to differ from the Company's forward-looking statements. These factors include inaccurate assumptions and a broad variety of other risks and uncertainties, including some that are known and some that are not. No forward-looking statement can be guaranteed and actual future results may vary materially. The Company does not assume the obligation to update any forward-looking statement. One should carefully evaluate such statements in light of factors described in the Company's filings with the Securities and Exchange Commission, especially on Forms 10-K, 10-Q and 8-K if any ; . In Item 1 of the Company's annual report on Form 10-K for the year ended December 31, 1999, which will be filed in March 2000, the Company discusses in more detail various important factors that could cause actual results to differ from expected or historic results. The Company notes these factors for investors as permitted by the Private Securities Litigation Reform Act of 1995. Prior to the filing of the Form 10-K for the year ended December 31, 1999, reference should be made to Item 1 of the Company's annual report on Form 10-K for the year ended December 31, 1998. One should understand that it is not possible to predict or identify all such factors. Consequently, the reader should not consider any such list to be a complete statement of all potential risks or uncertainties. In order to quantify effects on dye coupling, dopaminewas bath applied, and coupling within the entire dendritic tree of the injected cell was analyzed. However, local uncoupling due to transmitter releaseat singlemodulatory terminalsaffecting only single dendrites or even parts of dendrites is likely to occur under physiological conditions. Although dopamineis released predominantly from varicositiesand might reach its receptorsby diffusion over relatively large distances, dopaminergicterminals have also been shown to be involved in triadic synaptic complexes in both rodent Verney et al., 1990 ; as well as primate Goldman-Rakic et al., 1989 ; prefrontal cortex, suggesting highly localized effects of the transmitter. The distancebetweendopaminergic release sites and gap junctions, however, is not known to date, and whether dopaminereceptorsare located in close proximity to junctional complexes also remains to be shown. Since dopamine receptor-mediateduncoupling of gap junctions might occur via widespread"volume neurotransmission" i.e., undirected spreadof transmitter in a larger volume of cortical tissue; Lidow, 1995 ; or in a highly localized fashion, it is unclear whether activation of dopaminergic afferents uncouples an assemblyof neurons, e.g., an entire column, single neurons, or only local dendritic compartmentsfrom the functional syncytium. We have only analyzed the effects of exogeneoustransmitter on dye coupling without attempting to stimulate releasefrom dopaminergic terminals in the slice preparation. Dopaminergic fibers originating in the brainstemare cut during preparationand the diffusely projecting dopaminergic afferents are difficult to stimulate. The dopamine uptake inhibitor nomifensine did not reduce dye coupling, indicating that spontaneously releaseddopamine does not reach concentrationshigh enough to significantly affect gap junction coupling. Microelectrodes rather than patch pipettes were usedfor tracer loading of pyramidal cells to exclude false negative resultsdue to washoutof componentsof intracellular second messenger pathways. However, to obtain higher resolution recordings, we usedthe patch-clamptechnique to study the influence of gap junction closureson electrotonic potentials. Under theseconditions, forskolin, which most effectively reduced tracer coupling, exerted effects on electrotonic potentials in five out of nine neuronstested. The lack of effect in four cells might be due partially to a long electrotonic distance between the location of gap junctions and the recording site, although a dialysis of intracellular componentsmight also have taken place by the time PKA-mediated phosphorylation reduces junctional communication. Cyclic adenosinemonophosphate dopamineas negative and regulators of gap junctional communicationduring postnatal developmentof the neocortex In different tissues, PKA stimulation has heterogeneous effects on gapjunction coupling. Whereascoupling is enhanced some in non-neuronaltissues DeMello 1983; Saezet al., 1986; Burt and Spray, 1988; DeMello 1989 ; , an inhibitory effect of PKA activation hasbeen shown in the vertebrate retina Teranishiet al., 1983; Piccolino et al. 1984; Lasater and Dowling, 198.5; Lasater, 1987; DeVries and Schwartz, 1992; Hampsonet al., 1992; McMahon, 1994 ; . Our resultsdemonstratea suppressiveeffect of raising intracellular CAMP concentrations dye coupling in the on developing rat neocortex. This second-messenger pathway is regulated by a number of G-protein oupled neurotransmitter receptors, suggestingthat chemical and electrical communication systemsare closely interrelated in the immature neocortex.
T d o Fallot with absent pnlmonk valve TOFAPV ; is a rare but life-threatening syndrome. Children with this complex may have a n e dilated pulmonary arteries which compress the tracheobronchial tree resulting in airway compromise and death. We reviewed the records of all 10 children with TOFAPV who were seen at our institution from 1962 until present. This review showed that, in om hospital prior to 1980, there was 1 mortality in infants M less than 6 months of age who presented with marked respiratory compromise. Within the last. year, however, two infants with respiratory distress secondary to TOFAPV were treated differently. The diagnosis of TOFAPV and massive pulmonary artery dilatation was confirmed by catheterization. The children were placed in a semiprone position on a He&t board and received vigorous chest physiotherapy. Subsequent surgical treatment was performed wben the children were 8 and 11 months old. After closure of their ventricular septal defect and reconstruction of the rigbt ventricular outflow tract RVOT ; , the pulmonary artery aneurysms were resected Sections from the dilated pulmod nary arteries were histologidy normal. Pulmonic i ciency, mild RVOT gradients and reduction in pulmonary arterial dilatation were observed on cardiac catheterization 3 and 6 months postoperatively. Both children are growing and wen more than 8 months after tbeir operations. We conclude that the treatment for tetralogy of Fallot with absent pulmonic valve presenting in infancy with respiratory distress is aggressive medical intervention fonowed by early surgical repair including resection of pulmonary artery aneurysms. CONCLUSIONS 1. Laboratory and yard tests of corrosion initiation and propagation were conducted over a period of several years with regular production culvert pipes from 2 different manufacturers designated by O concrete made with unblended cement ; and F concrete made with cement blended with fly ash ; . Laboratory exposure for 4.4 years to cyclic saltwater ponding resulted in initiation of sustained corrosion in all three F specimens at times ti ranging from ~ 0.3 to ~3 years, but in none of the three O samples. Laboratory exposure for 3.4 years to continuous simulated seawater ponding resulted in sustained corrosion initiation for only one of three F specimens at ti ~ 2.6 years, but no initiation for the other two F or three O specimens tested. 2. The difference in corrosion initiation behavior between O and F does not necessarily reflect any special corrosion resistance of the type O pipe materials, as the O specimens tested had significantly thicker average rebar concrete cover than those of the F specimen group. 3. Surface-averaged corrosion rates measured in the laboratory specimens that showed sustained corrosion initiation were on the order of 1 m year to 10 m year. Potential map measurements suggest localization of corrosion with corrosion rates about one order of magnitude higher than average rates. No external manifestations of corrosion rust stains, cracks ; were detected to date in the ponding regions of any of the specimens from either supplier. 4. Chloride concentration profiles for the laboratory specimens were used to estimate that the chloride diffusivity for O and F concrete was on the order of ~10-7 cm2sec-1 and ~210-8 cm2sec-1 respectively. These diffusivity values are higher than commonly encountered in low permeability concrete, even though in both cases the water to cementitious ratio was very low. This behavior may be due in part to the modest cementitious content reported for both materials, and suggests that performance may be improved by the use of higher cementitious content concrete containing fly ash. 5. The critical chloride concentration for F was estimated to be between ~ 0.8% and ~1.8% of the total cementitious content, a range of values somewhat larger than those normally reported for steel in concrete. The critical concentration for O was estimated to be ~1.6% of the total cementitious content. These estimates are subject to considerable uncertainty and may be revised in continuing testing, which should also explore unresolved issues on chloride profile complexities and interpretation of concrete resistivity results. 6. Yard exposure in duplicate of full-size samples of O and F culverts was conducted by preconditioning without chloride contamination for ~ 2 years and then by partially filling the pipe with saltwater for ~2.2 years. Steel in O and F culverts reached long-term steady potentials of ~-300 mV and ~-600 mV CSE ; respectively. Apparent polarization resistance of the steel in the O culverts was several times greater than that in the F culverts. While interpretation of these results is not straightforward, these symptoms suggest that the steel in the F culverts had reached a condition of sustained active corrosion during the test period.
