DISCUSSION During exocytosis membrane components of chromaflin granules should become exposed on the surface of isolated chromaffin cells. We have studied this process in a quantitative manner using specific antisera against two membrane components. Both are glycoproteins having at least some of their antigenic groups on the inner surface of the chromaffm granule membranes. Dopamine , B-hydroxylase is the major glycoprotein of the membrane of chromaffin granules. A significant concentration of this enzyme 5% of total protein ; is, however, also found in the soluble lysate 31 ; . The other antigen, glycoprotein III, is the second major glycoprotein of the granule membrane 8, 9 ; . This glycoprotein is firmly bound to the membrane since it is not removed by washes with hypertonic solutions or with low concentrations of detergents 9 ; . In the soluble granule content only a very small concentration of glycoprotein III 0.25% of total protein ; is found 9 ; . Therefore, this protein is a more useful membrane marker than dopamine -hydroxylase . We thus focused our attention on glycoprotein III, although the experiments with dopamine 0-hydroxylase gave essentially identical results . Qualitative evaluation of immunofluorescence revealed both antigens to be present on the plasma membrane in tiny spots and or patches irrespective of the pretreatment of the cells. It seems unlikely that this patchy pattern was induced bythe reaction with antibodies causing aggregation of antigens originally randomly distributed in the plasma membrane . Antibody treatment was performed at 0 C the presence of sodium azide . This is known to inhibit active processes depending on intact cell metabolism, i .e., capping and subsequent stripping of membrane antigens 2, 20 ; , but does not prevent passive antigen redistribution in the membrane . However, the same patchy immunofluorescence appeared when cells were fixed with 1 .3% formaldehyde before immunostaining, which is known to preserve the original distribution of antigens in the membrane 30 ; . After stimulation the intensity of specific membrane fluorescence was observed to increase, whereas further incubation at 37C after stimulation reduced it again . However, it was difficult to ascertain these changes by conventional fluorescence microscopic observations, since apparently not all cells were affected by the stimulation to the same degree . To the Editor: It has been well established that cyclic 3', 5'-adenosine monophosphate cAMP ; is elevated in the cerebral cortex following anoxia, 1 ischemia, 2 trauma, 8 and decapitation, 4"8 and in the cerebrospinal fluid CSF ; in experimentally induced cerebrovascular disorders, ' in patients with acute cerebral contusion and concussion, 8 and in patients with stroke.'" Phenothiazine tranquilizers have been shown to inhibit the rise in cerebral cAMP following decapitation12 and injury, 3 without altering basal levels of cAMP. 12 In addition, one of the phenothiazines, chlorpromazine, has been shown to protect liver cells from death due to ischemia.18 ; Of the phenothiazines tested against a rise in cerebral cAMP, trifluoperazine most efficiently inhibited that rise, a dose of 20 mg kg inhibiting the rise by nearly 100%. For that reason, we have tested whether trifluoperazine would also have a protective effect in gerbils with experimental cerebral infarction. Trifluoperazine 20 mg kg ; in a buffer solution was given intraperitoneally to 31 animals 90 min before operation. Thirty control animals received an equal volume of the buffer solution and 42 control animals received no treatment. Infarction was produced by ligation of the left common carotid artery. The animals were evaluated every 8 hours for 88 hours in a double-blind manner. In the trifluoperazine-treated group, 25.8% 8 of 31 ; had infarcts, a lower incidence than in the buffer-treated group, of which 63.0% 17 of 27 ; p 0.005 ; had infarcts and in the nontreated group of which 51.2% 21 of 41 ; p 0.05 ; had infarcts. Mortality was not significantly lowered in the trifluoperazine group, but the trifluoperazine-treated animals had a lower probability of dying than the nontreated animals p 0.05 ; at every 8-hour observation period and a lower probability of dying than the buffer-treated animals p 0.05 ; at 6 of the eleven 8-hour observation periods. Thus, we believe that trifluoperazine prevented the small, nonfatal infarcts that were seen in the control groups. Since pentobarbital has been reported to inhibit elevations in cAMP and to offer a protective effect in experimental cerebral ischemia, 111 ' and theophylline has been reported to lower mortality in experimental stroke, 20 there may be a relationship between the prevention of an ischemia-induced rise in cortical cAMP and the lowering of morbidity and mortality in experimental stroke. Although trifluoperazine has not been used in the treatment of stroke, our results show that it provides a protective effect against cerebral ischemia similar to that seen with pentobarbital, and suggest a possible mechanism for pentobarbital protection. | Other Interesting Ericaceae Five other ericad genera Chimaphila, Kalmia, Moneses, Orthilia, and Pyrola are interesting but nearly impossible in cultivation. A number of species of the five are indigenous to western mountains, from California northward. All generally grow in moist coniferous woods, some at higher elevations. Kalmia species prefer bogs and moist to wet meadows, K. microphylla in higher mountains and K. polifolia at lower elevations, often coastal. Pyrola asarifolia, with its pink to rose flowers, and P. picta, white flowers and white leaf veins, are quite colorful. Orthilia secunda one-sided wintergreen ; is a neat 6-10-inch plant of moist woods in higher mountains, Chimaphila umbellata prince's pine or pipsissewa ; and C. menziesii little prince's pine or little pipsissewa ; are intriguing beyond their names. Both are little gems in coniferous forests, their pink to red flowers hanging downward in modesty at tips of stems. Through its common name, Moneses uniflora woodnymph ; speaks for itself. The Parasitic Ericaceae No native Ericaceae are more colorful or fascinating than those of the parasitic genera Allotropa, Hemitomes, Monotropa, Pityopus, Pleuricospora, Pterospora, and Sarcodes. Pyrola aphylla leafless wintergreen ; , actually leafless forms of P. asarifolia and P. picta, are sometimes included. Virtually all are uncommon to rare. All are found in open woods to deep forest, usually where moist, from California northward except as noted. These plants have one thing in common. All obtain nutrition from green plants, often conifers, through fungal intermediates known as mycorrhizae. Formerly they were thought to be saprophytes, but their parasitic nature has been rather thoroughly researched in recent years. Their common names are almost as fascinating as the plants themselves. The red and white striped Allotropa virgata candy stick or sugar stick ; is sweet to the taste. Hemitomes congestum gnome plant ; an inch-high folklorish dwarf, pink upon emerging, later aging straw color and to heights of 2-5 inches or more.
